چکیده

Likemostmatter (with the exception of, say, neutron stars), proteins and peptides are mostly made of nothing – an ephemeral cloud of electrons with very little mass surrounding tiny and very dense atomic nuclei that contain nearly all of themass (i.e., peptides and proteins aremade of atoms). Atoms havemass and the unit ofmass that is most convenient to use is called the atomic mass unit (abbreviated amu or u) or in biological circles a Dalton (Da). Over the years, physicists and chemists have argued about what standard to use to define an atomicmass unit, but the issue seems to have been settled in 1959 when the General Assembly of the International Union of Pure andApplied Chemistry defined an atomicmass unit as being exactly 1/12 of themass of the most abundant carbon isotope (C) in its unbound lowest energy state. Therefore, one atom of C has a mass of 12.0000 u. Using this as the standard, one proton has a measured mass of 1.00728 u and one neutron is slightly heavier at 1.00866 u. One C atom contains six protons and six neutrons, the sum of which is clearly more than the mass of 12.0000 u. A carbon atom is less than the sum of its parts, and the reason is that the protons and neutrons in a carbon nucleus are in a lower energy state than free protons and neutrons. Energy and mass are interchangeable via Einstein’s famous equation (E1⁄4mc), and so this “mass defect” is a result of the nuclear forces that hold neutrons and protons together within an atom. This mass defect also serves as a reminder of why people like A. Q. Khan are so dangerous [1]. Each element is defined by the number of protons per nucleus (e.g., carbon atoms always have six protons), but each element can have variable numbers of neutrons. Elements with differing numbers of neutrons are called isotopes and each isotope possesses a different mass. In some cases, the additional neutrons result in stable isotopes, which are particularly useful inmass spectrometry (MS) in amethod called

DETECTION OF CROSS-LINmD PERIDES BY FAST ATOM B01WBARDMENT MASS SPECTROMETRY

The possibility of chemical modification of peptides and proteins under the condition of proteolytic digestions and FABMS analysis was investigated. The results ;indicate that among the amino acid constituents of peptides and proteins: serinefcysteine, and cystine are the most sensitive residues which undergo chemical modificadons under the exprimenta1 conditions. The chemical modification ...

HIGH-PERFORMANCE LIQUID CHROMATOGRAPHIC ANALYSIS OF SULFUR MUSTARD REACTION WITH AMINO ACIDS AND PROTEINS

The interaction of sulfur mustard with aminoacids and proteins has been investigated in this study. Rats were injected with sublethal doses of sulfur mustard subcutaneously and intraperitoneally. At different time intervals, plasma and urine samples were collected. The binding affinity of sulfur mustard with urinary and plasma proteins and enzymes was studied for the first time using non-i...

Thermodynamic-Biochemical Study of Complexes of Intermediate Elements with α-Amino Acids in Some Proteins with Active Site

In this paper, the quantum chemistry calculations related to the structural parameter of the chromite and molybdate anions and the complexes obtained from them with the glycine and alanine amino acids were performed. The calculations were carried out using HF and DFT methods and in the base series 6-31G *. Thermodynamic studies related to the formation of complexes have been considered and thei...

Pattern of activity levels of amino trnasferases in the gonads of silkworm, Bombyx mori L.on exposure to prostaglandin F2

The impact of PGF2? on total proteins, free amino acids and the amino transferases in the gonads of silkworm, Bombyx mori L. were investigated in the present experiment. The effect of PGF2? resulted in increase in the total protein and free amino acid contents in the gonads of silkworm larvae indicating improved structural and dynamic levels of organization. The enhanced activity levels of amin...

Capillary electrophoresis-electrospray ionization mass spectrometry of amino acids, peptides, and proteins.

Separation in capillary electrophoresis (CE) is based on the movement of charged compounds inside a background electrolyte under an applied potential. Because the mechanism of separation of CE differs from that of conventional high-performance liquid chromatography (HPLC), where separation is based on the analyte's hydrophobic properties, CE is often used as a complementary technique to HPLC. I...

[Identification of amino acid phenylthiohydantoins by mass spectrometry].

The unequivocal identification of phenylthiohydantoins of the 20 amino acids usually occurring in proteins is possible by mass spectrometry and by combined gas chromatography-mass spectro metry. The method is sensitive enough to be applied to the identification of phenylthiohydantoins obtained from automated Edman degradations of polypeptides. Mass spectra of 21 amino acid phenylthiohydantoins...
